Isocitrate dehydrogenase 1 (IDH1) gene encodes a NADP+-dependent isocitrate dehydrogenase playing prominent roles in cellular metabolism including lipid metabolism and glucose sensing (1).

Somatic mutations have recently been identified in multiple cancers including early- and late-stage glioma (2-3), sarcoma (4-5), acute myeloid leukemia (AML; 6-7), and others. Notably, all mutations map to arginine residues in the catalytic pockets of IDH1 (R132).
Particularly the recent discovery of a gain-of-function somatic mutation in gliomas and its potential role in disease pathogenesis and prognosis has made IDH1 both a promising biomarker and potential therapeutic target (8-9).
