Estrogen receptor 1 gene encodes for a ligand-activated transcription factor localized in the nucleus which is involved in hormone binding, DNA binding, and activation of transcription.

Several different point mutations in the ligand-binding domain of ESR1 have been identified in tumor samples from patients with ER-positive metastatic breast cancer after treatment with anti-estrogen therapy, which may mediate anti-estrogen resistance (1-4).
